I put one of the Hill People Gear organizer panels (https://hillpeoplegear.com/Products/CategoryID/6) in a day pack I use. Added the "Tactigami" holster and mag carrier from Vertx, and have been very happy with the setup. Still not a quick access option, but certainly more so than a zippered case within a closed pack. You can mount the holster right at the top of the panel, just inside the pack opening. Stays put too. (Note the panels fasten right into the HPG packs. Probably would require some rigging on another brand.)
Maxpedition make some packs with dedicated pistol pouches.

Maxpedition used to have a Velcro insert that was adjustable at the trigger guard. I use that with a trigger guard cover like a Vanguard etc, lashed to the pack.
A RCS pocketshield could also work well in some compartments based on size or molle availability and that would allow you to used a dedicated kydex holster with it.
