Exactly how effective is it by touching the rear green vials with a red sharpie? I'm assuming it's not super difficult to wash off or remove if I don't like it?

I have no idea. Chuck Haggard is the one to ask about that. I've never seen a picture of it, but if someone has one please do post it.

It will probably come off easy with alcohol or acetone. In fact, removing it is not what I'm worried about; I'm worried about it coming off too easily, so I may need to escalate to some other clear red coating.

Red sharpie is effective, but pretty easy to remove IME. I think I'm going to try a dab of clear nail polish over it next time I touch up.

How bad is it if I just ordered a set of Operator G/Y for a Glock 43 that I don't even own, and am not sure I want to buy?

I'd call it being very self aware. We all know at some point you're buying a 43 and subconsciously you do too.
