"Are you ready? Okay. Let's roll."- Last words of Todd Beamer
I don't know about the jail, arrestee and trunk part, but i routinely remove the QLS mounted holster with the pistol still in it, to switch to another holster and pistol.

I do it on a daily basis, and I have the locking one as well.
I use two hands pinch inwards and out on each side and then lift it out.
I've been using one for about 5 years so I can't remember if it was difficult at first but its pretty much second nature to me at this point.
Just Like this picture, after field strip:
The slide stop is locking the take down safety to the rear, keep the sear down and keep the take down lever in position.
During reassembly, lock the slide back (pushing up the slide stop), will release everything. Not a problem to me.
Use a dremel remove some material from the slide stop or take down safety(yellow arrows),if you don’t like it. Choose a cheaper part to modify. 😂
Even you can dremel off the tip of take down safety, after that just pull the trigger to lower the sear when field strip. Just like a Glock. Take down safety can not save a careless gun user every time, you can not fix stupid.

IMO I am not too worried about recent P320 safeties except:
1)
Everyone knows this, the trigger itself physical inertia when dropped at a certain angel.
So use light weight trigger shoe, don’t use lighter trigger bar spring, no lighter sear spring.
2)
As the picture shows, the striker safety spring is small and light, the block interface is small.
If the sear and striker leg are accidentally disconnected during a heavy drop, the striker safety may not strong enough to stop the momentum of the striker itself. So at least no lighter striker safety spring, no lighter sear springs. Improve skills not the trigger.
